13-P061 Wnt signalling via Pitx2 regulates somite and limb myogenesis

چکیده
Several signalling pathways have been implicated in early liver development and mosaic analysis has revealed an essential involvement of the lateral plate mesoderm (LPM) in this process. Consistently, a zebrafish gene, prometheus/wnt2bb, bilaterally expressed in the LPM adjacent to the forming liver, is required for liver specification. Although liver specification initially fails to occur in prt/ wnt2bb mutant embryos, a small number of endodermal cells start expressing liver-specific genes at later stages, suggesting that additional factors act in parallel in liver specification and compensate for its absence. Studies in chick have shown a complex pattern of defined Wnt gene expression domains along the intestinal anlage. Intriguingly, we find zebrafish Wnt2, closely related to Prt/ Wnt2bb, expressed in an appropriate temporal and spatial window to represent a good candidate to act in conjunction with Prt/Wnt2bb in liver formation. Therefore, we tested its involvement in this process. While Wnt2bb/Prt promotes hepatoblast specification; we observe Wnt2 function is required for subsequent hepatoblast proliferation. These findings suggest a sequential requirement of Wnt signalling in liver formation. Intriguingly, double knockdown of both Wnt ligands revealed a cooperative role in hepatoblast specification. Excitingly, this highlights the complexity of a wnt signalling network in liver/endodermal organogenesis.

Pitx2 defines alternate pathways acting through MyoD during limb and somitic myogenesis.
The MyoD gene is part of the core regulatory network that governs skeletal myogenesis and acts as an essential determinant of the myogenic cell fate. Although generic regulatory networks converging on this gene have been described, the specific mechanisms leading to MyoD expression in muscles of different ontology remain misunderstood. We now show that the homeobox gene Pitx2 is required for in...
متن کاملExpression of (beta)-catenin in the developing chick myotome is regulated by myogenic signals.
The developmental signals that govern cell specification and differentiation in vertebrate somites are well understood. However, little is known about the downstream signalling pathways involved. We have shown previously that a combination of Shh protein and Wnt1 or Wnt3a-expressing fibroblasts is sufficient to activate skeletal muscle-specific gene expression in somite explants. Here, we have ...
متن کاملCell-autonomous integrin control of Wnt and Notch signalling during somitogenesis.
Integrins act at signalling crossroads, and their interactions with other signal transduction pathways are key to the regulation of normal and pathological cell cytoarchitecture and behaviour. Here, we describe a signalling cascade that acts during the formation of the defining segmental features of the vertebrate body - the somites - in which β1-integrin activity regulates epithelialisation by...
متن کاملGli2 and Gli3 have redundant and context-dependent function in skeletal muscle formation.
The Gli family of zinc finger transcription factors are mediators of Shh signalling in vertebrates. In previous studies, we showed that Shh signalling, via an essential Gli-binding site in the Myf5 epaxial somite (ES) enhancer, is required for the specification of epaxial muscle progenitor cells. Shh signalling is also required for the normal mediolateral patterning of myogenic cells within the...
متن کاملThe ADAMTS5 Metzincin Regulates Zebrafish Somite Differentiation
The ADAMTS5 metzincin, a secreted zinc-dependent metalloproteinase, modulates the extracellular matrix (ECM) during limb morphogenesis and other developmental processes. Here, the role of ADAMTS5 was investigated by knockdown of zebrafish adamts5 during embryogenesis. This revealed impaired Sonic Hedgehog (Shh) signaling during somite patterning and early myogenesis. Notably, synergistic regula...
